Cytokeratin 6 antibody clone LHK6 is a monoclonal antibody that recognizes cytokeratin 6, a type II intermediate filament protein expressed in keratinocytes of stratified epithelia. Cytokeratin 6 is normally present in the outer root sheath of hair follicles, the nail bed, and certain mucosal tissues. It is also upregulated in conditions of hyperproliferation, tissue repair, and carcinogenesis, making it a versatile marker for epithelial biology. NSJ Bioreagents offers Cytokeratin 6 antibody clone LHK6 for researchers studying epithelial growth, wound healing, and tumor pathology.
Cytokeratin 6 antibody clone LHK6 produces cytoplasmic staining in keratinocytes and has been widely applied in dermatopathology. It is frequently used to identify squamous cell carcinomas, where cytokeratin 6 expression is often elevated. The antibody also labels basal keratinocytes in reactive epidermis, making it a reliable indicator of epithelial activation. Pathologists use cytokeratin 6 antibody clone LHK6 alongside other keratins to classify tumors and determine their tissue of origin.
In wound healing, cytokeratin 6 is rapidly induced in keratinocytes migrating into the wound bed. This expression facilitates epithelial repair and barrier restoration. Cytokeratin 6 antibody clone LHK6 allows researchers to monitor this process in experimental models of skin injury, providing insight into the dynamics of keratinocyte migration and proliferation.
In addition to skin biology, cytokeratin 6 antibody clone LHK6 has applications in tumor research. Overexpression of cytokeratin 6 has been observed in head and neck squamous carcinomas, cervical cancers, and lung cancers. Its detection contributes to tumor classification and can provide information about tumor aggressiveness. The antibody is also used in studies of epithelial stem cells and differentiation pathways, reflecting its broader role in epithelial regulation.
Cytokeratin 6 antibody clone LHK6 has been validated for immunohistochemistry, immunofluorescence, and western blotting. It consistently produces strong cytoplasmic signals in positive cells and has been cited in numerous publications focusing on epithelial differentiation and pathology. Alternate names include CK6 antibody, keratin 6 antibody, and keratin type II cytoskeletal 6 antibody.
The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the antibody to be titered up or down for optimal performance.
1. Staining of formalin/paraffin tissues is enhanced by digestion of tissue sections with pepsin at 1mg/ml Tris-HCl, pH 2 for 10-15 min at RT or pH 9 10mM Tris with 1mM EDTA for 20 min at RT.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
A synthetic peptide of 11 amino acids (GSSTIKYTTTS) from the C-terminus of human Cytokeratin 6 was used as the immunogen for this antibody.
Store the Cytokeratin 6 antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
